U-CyTech biosciences switches from roller bottles to FiberCell Systems Hollow Fibre Bioreactor

U-CyTech biosciences is based in Utrecht, the Netherlands, and produces a wide range of innovative cytokine ELISA, ELISPOT and Fluorospot systems (human, monkey, rodent). Productions of monoclonal antibody with hybridomas are now performed at U-CyTech’s Utrecht laboratory using the FiberCell Hollow Fibre Bioreactor system. This is a compact system for Hollow Fiber cell culture and mAb production.

Previously, U-CyTech produced mAb’s in roller bottles. However, this method was labor intensive, time consuming and a lot of medium was needed. Therefore, a permanent switch to the method of continuous, high density hollow fiber cell culture was made using the FiberCell System.

Monoclonal antibody - SDS gel - raw supernatant

Clone 1: 517 ml raw supernatant. Centrifugation, filtration, AS precipitation, dialysis, ion chromatography, 0,22 mM filtration. Yield >95% pure IgG
526 mg IgG in 10 weeks

Gel SDS FiberCell CDM-HD

Clone 2:  543 ml raw supernatant. Centrifugation, filtration, AS precipitation, dialysis, ion chromatography, 0,22 mM filtration. Yield >95% pure IgG
1029 mg IgG in around 10 weeks.

The left hand lanes of the above SDS gels show diluted raw supernatant. The media used was DMEM+CDM-HD + Glutamine 2 mM (1/100 Gibco 25030 200mM)+ PenStrep 1/100 (Gibco 15140). CDM-HD is recommended by FiberCell Systems for use with their bioreactor. With this system, U-CyTech has been able to simplify the purification procedure:

“… purification after culture is now shortened and improved. The concentration step, ultrafiltration, is not necessary with the Fibercell (We used to have yield loss during concentration). Now we only perform an ammonium sulfate precipitation and a chromatography step. The ion chromatography is more efficient because after AS precipitation the Fibercell material is much cleaner than from the roller bottles after AS precipitation.”  – U-Cytech
