KDK096 SP5D 96-Well Plate (25 spheroids per well)
Patient derived tumour spheroids provide a revolutionary tool that opens the way to designing cancer treatments tailored to the unique profile of individual patients. The new SP5D 96 well plate provides 12 strips of 8 wells with 25 microwells per well for spheroid formation, i.e. 2400 spheroids in total, increasing the number of replicates without extra pipetting. The removable well strips (200 spheroids) can be used individually.
It is the ideal product for accelerating the pace for time-critical cancer projects.
In SP5D spheroid culture plates the microscopic wells are produced with a hemispherical base to drive the rapid clusturing of cells into 25 rounded spheroids per microwell. A unique nanocoating prevents cells from adhering to the surface. Consistent spheroids are formed, and in a single macrowell the spheroids share the media, fostering more natural communication between cells.
The new SP5D 96 Well platform is ideal for easily generating 25 spheroid replicates per well thereby boosting the statistical relevance of data from patient derived tumour samples.
Preparation
Prepare functional well with 0.1 mL of medium

Addition of Cells

It has been shown (1) that iPSC derived multi-tissue organoids can be grown for up to 30 weeks in G-Rex bioreactors which provide a continuous exchange of gas at the cell/tissue interface via a basal gas-exchange membrane. Little routine maintenance is required apart from refreshing media every few days. G-Rex culture systems are produced to GMP grade by WilsonWolf Manufacturing LLC. and represent the gold-standard culture method for the rapid expansion of T-Cells destined for CAR-T therapy, for example (2).
G-Rex technology is available in research tool format : 6- and 24-well multi-well plates. See product page.

In this example, here are the results of the development of monoclonal antibody for 10 newly developed monoclonal cell lines.
Following the successful fusion, clone selection and weaning from media supplements, candidate clones were transfered to wells of G-Rex 6 Well Plates (35ml media per well, and 10cm2 gas exchange membrane per well).
The cell culture supernatant was purified with protein A. The objective here was to obtain initial quantities of purified antibodies for comparison purposes rather than to try and optimize the culture conditions. G-Rex is the perfect device for the development monoclonal antibody since each well acts as an individual “mini-bioreactor”, which is self-sustained due to G-Rex’s gas exchange membrane technology.

G-Rex Plates enable large numbers of any type of non-attachment cell to be grown to high numbers within a few days: hybridomas, THP-1, CHO, HeLa, SF9 etc.
In a G-Rex 6 Plate 5x10e6 cells can be expanded to between 200 – 400 x10e6 within around 10 days with only one exchange of medium. Hybridomas can produce high-titer antibody in useful amounts after 10 to 14 days.
A gas permeable silicone membrane at the base of each well ensures efficient oxygenation during cell expansion. This results in exponential growth of cells limited only by the amount and type of media in the well. Passaging is no longer required and high cell numbers are achieved with minimal effort.
G-Rex Plates free up space for other work in an incubator so can make a valuable contribution to the efficiency and productivity of many cell culture labs.
