Scale-up Production of Conditioned Medium / Secretome Generation

The potential regenerative or immunomodulatory applications of secreted factors from cells in culture or their relevance as disease biomarkers is driving interest in the use of more efficient ways to collect enriched conditioned media. A compact FiberCell® Hollow Fibre Bioreactor provides an ideal way to produce large amounts of cell-conditioned medium, and to generate an enriched secretome, including extracellular vesicles from various cell types such as mesenchymal stem cells. Continuous perfused culture of up to 1E+11 cells coupled with the use of 5Kd or 20Kd MWCO hollow fibres enables periodic collection of media containing highly concentrated cell-secreted factors. This process can be continued over many weeks and only requires the space of a shelf in a standard incubator.

Optimize your production run by choosing between a 5 kd or 20 kd molecular weight cut-off fibre.

Recent studies have shown that apoptosis is significantly reduced in the hollow fiber environment. The supernatant is free of intracellular proteins that might come from cell lysis.

A FiberCell® Bioreactor, complete with media reservoirs, will easily fit inside a standard incubator.

In high density cultures cell-secreted factors will support growth to an extent that serum can be replaced by a chemically-defined protein-free, xeno-free formulation, CDM-HD.
